چکیده
Abstract Background There is great interest in utilizing nanoparticles to improve drug delivery central nervous system (CNS) tumors, but the biologic features underlying successful at tumor site are not known. METHODS We developed a pooled screening assay investigate cellular predictive of nanoparticle and screened 35 fluorescent formulations against 488 cancer cell lines with DNA barcodes. The library was comprised nonlethal liposomal or polymeric cores varied surface functionality, including natural synthetic polymers. Cells were profiled using fluorescence-activated sorting barcodes sequenced deconvolved generate an association score for each nanoparticle-cell line pair. RESULTS Of lines, 22 lineages (tissues origin) included. derived from CNS tumors; these classified as astrocytoma glioblastoma, one oligodendroglioma, 2 medulloblastoma. 40% female patients average age derivation 52 years; TP53 hotspot mutations detected 25/35 PTEN 15/25 lines. Using unsupervised hierarchical clustering by profiles, did cluster together lineage, subtype, mutation status rather baseline expression genes involved nanocarrier trafficking. Nanoparticle profiles heterogeneous among most formulations, though had high polystyrene respect other lineages. Select uptake liposomes coated polymers fucoidan, alginate, hyaluronic acid chondroitin sulfate; trends further probed non-pooled screen 5 additional Conclusions Uptake across can be predicted gene expression.

Generation of Cisplatin-Resistant Ovarian Cancer Cell Lines
Ovarian cancer is the most lethal gynecological cancer in which cisplatin-based treatment plays fundamental role as the first line chemotherapy option. However, development of platinum-resistance is a critical and poorly understood problem in ovarian cancer treatment. Although in vitro generation of platinum-resistant ovarian cancer cell lines is a long established approach to uncover the molec...
متن کاملReporting the Effect of Cell Seeding Density on Growth Pattern of Cancer Cell Lines
In vitro studies on cancer cell lines still constitutes a major part of cancer research. It is very important that the researchers consider the importance of basic conditions of working with different cell lines. To the best of authors knowledge, this study is the first one to report the effect of seeding density on the growth curve of three human cancer cell lines. These cell lines include hum...
متن کاملCytotoxic and Cell progression effects of Mentha pulegium L extract on selected cancer cell lines
Mentha pulegium L. is one of the Mentha (Labiatae) species that is grown in many different parts of the earth including northern Iran. The total ethanolic extract of the aerial parts of this plant is used in this work to investigate on the possible cytotoxicity on two different cell lines. Human lung carcinoma cells (A549) and human breast cancer (MCF7), have been used to show the clonogenic co...
متن کاملCell Kinetic Study of Tamoxifen Treated MCF-7 and MDA-MB 468 Breast Cancer Cell Lines
Apoptosis could be a major mechanism of antitumor effect of tamoxifen. Therefore this study is designed to characterize the kinetic behavior of tamoxifen-induced apoptosis in the estrogen receptor positive (ER+) and negative (ER-) cell lines, MCF-7 and MDA-MB-468. Frequency of cell death was examined by trypan blue and acridine orange staining. Annexin V-Fluorescein/PI was used in flow cytometr...
متن کاملAnticancer properties of chitosan against osteosarcoma, breast cancer and cervical cancer cell lines
Background: Cancer refers to the abnormal growth of cells and is still the most common cause of morbidity in world. The purpose of this study was to determine cytotoxicity effect of high molecular weight (HMWC) and low molecular weight of chitosan (LMWC) on three cancerous cell lines MCF-7, HeLa and Saos-2 with different histological origin. Methods: The anticancer property of two types of chi...
